Abstract:
objective: to shape hyperuricemia animal model similar to human metabolism and observe the effect of chicory extract on it. Methods: The quail was selected as a model animal, and the yeast bait was used as a modeling agent to shape the hyperuricemia model, and the serum uric acid and other biochemical indexes of quail were dynamically detected, to observe the effect of chicory extract on the serum uric acid level of the model animal. Results: the serum uric acid level of quails fed with yeast (15g/kg) for 7 days significantly increased for 5 weeks. A small dose of chicory extract administered for 7 days can significantly reduce the yeast-induced blood uric acid rise in quails and fight against the transient increase in blood sugar, and the effect of administration for 14 days is better than that of the positive control drug benzbromarone. Conclusion:(1) the model is an ideal animal model for screening traditional Chinese medicine for prevention and treatment of hyperuricemia and studying its mechanism. (2) chicory extract has obvious effect of lowering uric acid. (3) the mechanism of modeling and the mechanism of drug action need to be further studied.

Abstract:
objective to cultivate chicory (Cichorium endivia L.) it is a kind of chicory of Asteraceae, which has been proved to have certain antioxidant activity, but its application and antioxidant components are rarely studied. In this study, the antioxidant activity of cultivated chicory was systematically studied. Methods to extract and separate the cultivated chicory, screen the antioxidant active parts, and further study the antioxidant effect of the effective parts in vitro and in vivo. In addition, the preliminary safety evaluation was carried out, and the chemical components of the effective parts were isolated and identified, and the antioxidant activity was detected. Results The antioxidant activity of 60% ethanol eluate (KJ-I) in the extraction and separation process of cultivated chicory was significantly better than that of other extracts by FCR,ORAC and DPPH methods. In addition, KJ-I can effectively prevent the hemolysis of rabbit red blood cells induced by AAPH, suggesting that it can protect the body from oxidative stress injury. Protected by KJ-I tert-butyl hydroperoxide (t-BHP), hydrogen peroxide (H_2O_2) and 2,2 & prime;-azobis (2-methylpropylimidazole) dihydrochloride (AAPH) the experimental results of HepG2 cells, which caused oxidative damage and effectively reduced the level of reactive oxygen species (ROS) in damaged HepG2 cells, demonstrated that KJ-I had a protective effect on liver at the cellular level. In the experiment of acute liver injury induced by t-BHP in mice, Pathological observation showed that KJ-I can prevent the necrosis of liver tissue in damaged mice, while KJ-I can effectively reduce the levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum of damaged mice, reduces lipid peroxidation in liver tissue, increases reduced glutathione (GSH) levels in liver tissue, and increases superoxide dismutase (SOD) and glutathione-S-transferase (GST) in liver tissue activity, the above results further illustrate that KJ-I has the function of liver protection in in vivo experiments. Acute toxicity test and Salmonella typhimurium reverse mutation (Ames) test showed that KJ-I was safe and nontoxic. KJ-I was further isolated and the structures of the obtained compounds were identified. The antioxidant activities of these components were compared by ORAC and DPPH · scavenging ability test. KJ-I known ingredients include: kaempferol, kaempferol-3-o-β-d-glucoside, phenylethyl glucoside, adenosine, and the new compound 2-furfuryl alcohol-(5 & prime;& rarr;11)-1, 3-cyclopentadienyl [5,4-c]-1H-misoline. Among them, kaempferol has the strongest antioxidant activity, which may be the main component of the antioxidant activity of cultivated chicory. And 2-furfuryl alcohol-(5 & prime;& rarr;11)-1, 3-cyclopentaproz [5,4-c]-1H-misoline is a relatively rare derivative of the natural plant. Conclusion The extract KJ-I of chicory has a good antioxidant effect, and the most effective antioxidant component may be kaempferol. At the same time, KJ-I has a good liver protective effect. In addition, preliminary toxicity tests suggested that KJ-I was safe and nontoxic. Therefore, KJ-I may have potential development value in Food antioxidants and liver protective agents.
